The Importance of Biodistribution Analysis

The RDC biodistribution analysis is an essential test of the quality control to identify defects in the quality of RDC, which is usually the first indication when something might be wrong, found during an imaging procedure, especially because RDC is a novel drug for the treatment of cancer. Biodistribution study in animal models must be performed during the development of a novel radiopharmaceutical by research institutions, as an essential part of the quality assurance procedure, before clinical research on humans and laterally released for sale.

How to Perform Biodistribution Analysis

The biodistribution analysis is conducted in animals with normal phenotypes but more are being performed in animals with specific tumors by imaging the animals on either PET (Positron Emission Computed Tomography) or SPECT (Single-Photon Emission Computed Tomography) after RDC injection to measure the distribution of the radioactivity in tissues by counting or autoradiographic techniques.

For example, somatostatin receptors (SSTRs), which are overexpressed on the neuroendocrine neoplasms (NENs), can be identified by somatostatin analogs (SSAs). To evaluate the new pan-somatostatin receptors (pan-SSTRs) probe PA1, the biodistribution analysis is conducted to compare the performance between novel [64Cu]NOTA-PA1 and already discovered [64Cu]DOTA-TATE. The micro-PET imaging results of [64Cu]NOTA-PA1 and [64Cu]DOTA-TATE in A549 tumor-bearing mice show that both the tumor concentration and the tumor uptake of [64Cu]NOTA-PA1 are much higher than [64Cu]DOTA-TATE, suggesting that [64Cu]NOTA-PA1 has better performance than [64Cu]DOTA-TATE in tumor mouse models.
